Overview
The Fluorescence Staining Kit is intended for use in qualitative histological procedures to facilitate the fluorescent microscopic detection of Acid Fast Bacteria(AFB) and Tubercle Bacilli. The kit employs Auramine-Rhodamine dye, which binds to the lipoid capsule of acid-fast organisms, enabling their visualization under a fluorescence microscope. This staining method highlights these bacteria due to their resistance to decolorization, allowing for clear and distinct observation.
The fluorochrome stain, Auramine-Rhodamine, used in this staining method binds specifically to the mycolic acids present in the cell walls of acid-fast organisms. This binding creates a stable complex that is resistant to decolorization by acid-alcohol, commonly used in acid-fast staining procedures. The phenol component in the stain facilitates its penetration into the cells, even in the presence of decolorizers. For an organism to be classified as acid-fast, it must be able to withstand decolorization by acid-alcohol.
Following the initial staining, a counterstain, Potassium Permanganate, is applied to highlight the stained organisms, providing a contrasting background that enhances visualization under a fluorescence microscope.
