Overview

The GRAM Staining Kit is intended for performing Gram staining, which facilitates the initial classification of bacteria into Gram-positive and Gram-negative categories. This staining protocol aids in the preliminary identification and differentiation of bacterial types.

The Gram stain differentiates bacteria based on their cell wall properties. When stained with Crystal Violet, all bacterial cells form a Crystal Violet-Iodine complex. During the decolorization step, Gram-positive bacteria retain this complex due to their thick peptidoglycan layer, whereas Gram-negative bacteria lose the complex because their thinner peptidoglycan layer is more permeable. The Gram-negative bacteria then take up the counterstain. The effectiveness of the Gram stain relies on the integrity of the bacterial cell wall; if the cell wall is disrupted, it can affect the staining outcome and potentially lead to inaccurate classification.
