Overview

The Ziehl-Neelsen/Kinyoun Staining Kit is designed for qualitative histological procedures to visualize Acid- Fast Bacteria and Tubercle Bacilli under optical microscopy. The kit employs Carbol-Fuchsin, which penetrates the lipoid capsule of acid-fast organisms, allowing them to resist decolorization and be clearly detected.

The Ziehl-Neelsen (Kinyoun) staining technique utilizes Carbol-Fuchsin dye, which binds to mycolic acids in the cell walls of acid-fast bacteria. This dye is resistant to decolorization by acid-alcohol, allowing it to remain in the cells despite the application of a decolorizer. The phenol in Carbol-Fuchsin facilitates dye penetration into the cell wall, even after exposure to decolorizers. To confirm that an organism is acid- fast, it must retain the Carbol-Fuchsin dye despite decolorization. A counterstain, Methylene Blue, is then applied to provide contrast and highlight the stained acid-fast organisms.
